Design Notes
The DNA binding site was first screened for using BLASTn. All Zinc Finger Proteins created by Duke iGEM 2011 had binding sites that occurred 2 times or fewer in the E. coli genome. Binding sites for each zinc finger were first found via BLASTn screening and then entered into the program ZiFit, set to CoDA analysis, in order to generate the DNA coding sequence for each zinc finger protein. These coding sequences and their respective binding sites were synthesized de novo.
